To give 1) an introduction to modern methods of liquid chromatography with particular emphasis on reverse-phase high performance liquid chromatography (RP-HPLC), 2) an understanding of the various means by which RP-HPLC separations can be controlled, and 3) an understanding of the ways RP-HPLC separations in the literature have been designed.
Overview of liquid chromatographic methods (affinity chromatography, gel filtration chromatography, ion exchange chromatography, hydrophilic and hydrophobic interaction chromatography, and RP-HPLC; the theoretical background for chromatographic methods; mass transfer and diffusion; stationary phases; the potential of modifying the surface of porous silica; instrumental design; laboratory course on RP-HPLC). The course is an expanded version of the laboratory course in Physical Biochemistry.
